    1. VHL nt. 406 T-->G

      A 9 year-old boy was admitted to the hospital with generalized seisures and marked hypertension (110 bpm). Skeletal muscle hypertrophy, extraadrenal pheochromocytoma, a spinal cord tumor, and paraganglioma were all present in this patient. Leading to his diagnosis of VHL, the patient was found to have a mutation at c.193T>G, reported in the paper as p.S65A (c.406T>G) due to a difference in transcript sequence. DNA of peripheral blood cells was tested and DNA analysis of the VHL gene was performed. This was also done in the mother who was found to be negative, however the father’s DNA was not available for testing, therefore cannot definitely differentiate between an inherited or a de novo mutation in this patient. Common VHL manifestations of hemangioblastoma or retinal capillary hemangioma were not present. 16 months after his first admission, the patient is in good health. ACMG evidence codes: 'PP4' since the patient presents phenotypes highly specific for a disease with a single genetic etiology.
