c.293A>C (Novel)p.(Tyr98Ser)
150 index patients with pheochromocytoma/paraganglioma were evaluated. Phenotypic data were collected and germline mutations in five susceptibility genes (RET, VHL, SDHB, SDHD and SDHC) were tested. Out of 30 patients with familial presentation, 13 were clinically diagnosed as VHL syndrome. Using PCR and Sanger sequencing through peripheral blood, genetic testing confirmed a mutation at c.293A>C of the VHL gene in two unrelated probands. A 15 year old Asian Indian male presented with unilateral pheochromocytoma, sympathetic paraganglioma, renal and pancreatic cysts. Family history was positive. The second was a 31 year old Asian Indian female who presented with unilateral pheochromocytoma, CNS hemangioblastoma, and a pancreatic neuroendocrine tumor. Family history was also positive.
