53 patients with a molecularly confirmed VHL mutation (by PCR through EDTA-anticoagulated blood) and a pancreatic neuroendocrine tumor (ICT) were collected from the German NET-Registry and the German VHL-registry. An additional 296 patients were stated in this paper with a VHL associated germline mutation, but without pancreatic neuroendocrine tumors. 1 patient was found to have the above mutation (c.467A>G). They were a VHL patient with an ICT (pancreatic neuroendocrine tumor) taken from the VHL-registry. Other clinical manifestations outside of a pancreatic neuroendocrine tumor are possible, but were not discussed. ACMG evidence codes: 'PP2' because they observe a missense variant in a gene where missense variants are a common mechanism of disease.

    1. VHL Y156C

100 patients (age: 13–95 years) who had either one or two non-syndromic pheochromocytoma without a positive familial history, without any other endocrine tumours, without any clinical signs for von Hippel–Lindau disease, MEN type 2 or neurofibromatosis type 1, were analyzed. Two patients in the study (Patient number 1 and 5) were found to both have a germ line mutation in the VHL gene, p.Tyr156Cys. The specific nucleotide change was not specified. Genetic analysis was conducted in search of a germ-line mutation in VHL by PCR-sequencing on both strands of DNA. Patient 1 was a 13-year old male who had a bilateral pheochromocytoma and Patient 5 was a 43-year old female who had a unilateral pheochromocytoma.

  2. Oct 2026
    1. Screening of 92 unrelated patients with VHL disease revealed 61 DNA variants. No variants were found in 96 control cases. Direct sequencing on peripheral blood was systematically performed on exon 1 PCR products. Southern blotting was used to search for larger alterations to the gene. The Y156C (c.467A>G) missense mutation was found in a VHL type 1 (no pheochromocytoma) patient (patient no. V265). One unaffected family member was negative for this mutation.

Data was collected from 82 VHL mutation carriers in the Dutch VHL surveillance program. Seven patients were found with this missense variant in the VHL gene. The ages of the patients was represented as a range 7-88 (mean 45). Phenotypes were grouped together: pheochromocytoma and renal cysts, although it is unclear if only one of the seven patients had renal cysts. Family history was not described. This cohort was published later in PMID 31087189.

    1. Case 12In 1993, a 39-yr-old female with recent onset of hypertension was diagnosed with a right pheochromocytoma. At age 36, a suspected left pheochromocytoma was noted by CT and increased urinary metanephrines was confirmed at age 39 by [123I]MIBG scintigraphy. This imaging showed in addition an uptake located in the neck. MRI confirmed a left adrenal tumor and a left 2-cm vagal paraganglioma that was removed and confirmed histopathologically. Genetic testing identified the VHL p.Tyr156Cys mutation. No other tumor of the VHL spectrum was discovered. Three of her four sons were found to carry the mutation. 2. VHL p.Tyr156Cys

Twelve patients with hereditary non-SDHx head and neck paragangliomas were analyzed. Eleven of the 12 patients were found to carry germline mutations in the VHL gene. Blood was used for molecular genetic analyses and the NM_000551.2 reference sequence was used. The 39 year old female proband was found to harbour the p.Y156C (c.467A>G) mutation of the VHL gene. She presented with recent onset of hypertension and was diagnosed with a right pheochromocytoma. At age 36, a suspected left pheochromocytoma was noted by CT and increased urinary metanephrines was confirmed at age 39 by MIBG scintigraphy. This imaging showed in addition an uptake located in the neck. MRI confirmed a left adrenal tumor and a left 2cm vagal paraganglioma that was removed and confirmed histopathologically. No other tumor of the VHL spectrum was discovered. Three of her four sons were found to carry the mutation and all underwent clinical screening. One of these sons presented with a 2.4cm abdominal paraganglioma.
