This paragraph is quite deep and striking due to the way the writer elaborates and analogizes a sinner in the eyes of God and the Church. There is definitely passion in his words as he describes how sinful individuals could have potentially been during this time. To read the passage, it seems like some kind of a warning. I interpret it as one needing to take head to sinful actions as God see's all that you do and he judges your actions daily. Therefore, if he chooses not to allow you to wake the next day be mindful your actions on Earth could potentially lead you to the gates of hell. Now that he has established why so many are damned and what horrors await them... it is time for redemption. A fitting portion to any sermon, a chance to be saved and if you pass today. "remarkable vengeance to others..." This leaves a powerful psychological impression on the crowd to walk out on. It fills the individuals with thoughts and second guesses, "I thought I was devout and saved... but is this my day of vengeance?". I would be very curious to see what the crowd looked like on exiting, how many were tearful with shame vs holding their head high with the belief the sermon was about all those other sinners...

Reading this sermon really put an image in my head of how the congregation probably looked at the time he was preaching it. I honestly would have left and went home and prayed about it because I would have been so frightened that I was one of the ones going to hell. It's like he was scolding a child. Honestly I would have never returned back to this church. We all are sinners but the good thing about that is we have a forgiving God. He forgave us for all of our sins and for that I am merciful to his name.

He is letting them know how God is the head of everything just in case you did not know. You have what you have because of God including your health and wealth. You didn't get this on your own. This is His grace on you. God is keeping you and carrying you and what you have because of God. Then he tells them that these earthly things are nothing. God gives and God takes away. After reading Washington's, "Altanta's Compromise", it became more clear to me that although he championed for progress for the African Americans, he did not completely fought for their equal rights. It seems that White men had influenced him greatly and the feelings of inferiority that was instilled in him growing up still lingered inside him. This was evident because of how he wanted the freedmen to have success but he also encouraged them to continue to keep their head down and work hard for the White men. He also did not encourage them to fight for their right to vote. Dubois however, saw that more needed to be done to in order to actually see progress in the African American's lives. He felt they also needed to hold some power where they can influence change regarding their rights and doing industrial work only benefits the White men. It may have been difficult for many African Americans to decide who they would follow.

Booker pinpointing the South White people here and reminding them how his people who were slaves work for them without complaints and any strikes. He is reminding them that how his people’s hard work helped their land grow and take care of their parents and children. He is also giving them promise here that his people are going to take care of them in the future as they did in the past. At one point, I felt like Booker considered Negroes lower than South white people.

Tawanda mentions how her parents were separated from the city of their dreams by the "theater stage," not by Siberia. This could be an example of the rapid change in transportation and immigration over generations, and how borders can be confusing and in flux.

This imagery is easy to visualize because the writer describes the way he moves towards the bed and what he can see. I describes the things he's doing and what he's thinking.

These lines show the connection between the sister and brother. Further in the stories it shows memories of how the two share about things they would not say out to others.

The author uses sensory details to display imagery. These details specifically appeal to the eye. Describing the room with details like the shattered window and the records on the floor allow the reader to create a picture in their head of what the room looks like. This creates meaning because the reader gets a better picture of the scene. He/she can better understand the character as a whole.

Imagery is one of the literary techniques that help the author create meaning. This description allows the reader to imagine the position the narrator is in. The word "laced" is a vivid word used by the narrator to create a more human experience for the readers.

I enjoy the narrator/author's use of humor to lighten the mood during these moments. How often are we placed in weird, uncomfortable situations where we sometimes think of our life as a "traumedy".

This talk of political authority reminds me of an idea in philosophy. The term 'philovictor' is used to describe someone who argues only to be right, not to learn. Other comments are more offhand, dismissive - 'Nonsense.' 'Please! ' 'HA! ! ' - that kind of thing. This is a really strange note, but this makes me think of "Last Christmas." "I gave you my heart but the very next day, you gave it away." Shakespeare is talking to the one who received his heart from his love, but I read this line and that's the first thought that popped into my head.

I'm interested in if this disclaimer serves as an assurance that the reader interprets Catherine as intended, or she's eventually to turn what we're supposed to know about her on its head.

This part of the story shows imagery by describing A forty year old man that his been in prison for 15 years.

This shows a change of heart in the banker since he realizes that he has stolen more life from the lawyer than he thought of by the bet.

This quote shows that the banker can not stand the fact that the lawyer may win since he is of younger age than the banker.

I grew up in a Christian home, and through out my childhood, the Bible had always been more or less the same book as the Hebrew Bible, not many things distinguished the two books in my head. I knew that we had some differences such as the New Testament, but it never went much further in developing those thoughts to actually think of them as being two different Bibles. Let alone being interpretative in different contexts.

This shows that you care about the mental health of your students and want them to be in a good head space to do well in your class, as well as college in general.

I am having a hard time wrapping my head around what I need to do for my job as as a teacher already- Mandates are coming at me rapid fire without any model of how to meet the expectations. My first week back to teaching is insane. I have emails coming at me 24/7. Going off of the literal physical aspects of the painting, the line style is smooth with a "fuzzy" painting style, to create a vague amount of detail, as expressed by the horses' lack of eyes, and less emphasis on the woman's dress. In the scene the painting captures, a man and woman, presumably a couple, horseback riding as the focus of the painting. The horse even has a violin underneath its head. In the left corner behind the male figure, there is a man seemingly playing a violin. Near the horses legs, there are crowds of people/animals who also appear to be listening to music with glee. With the presumed couple as the main focus, it creates almost a sense of liberation. Although there are these people in the background, the woman is still topless wrapped in the arms of this man, uncaring of what others may think.

the author described this so well that I am able to see an image in my head of what's going on in this sentence. its helped me understand what exactly was going on when these two girls were killed.

You must first need clear intention of the problem that needs to be solved, and it has to be communicated clearly to teammates in order to head in the right direction. Without that, your team may take unnecessary detours.

In this part of the article there is lot of imagery used. From using words like Nutcracker ballet black leopard and twisted like a seashell. These terms show imagery and make the reader imagine it in their head.

I've heard that working in a public school is less work than working in a private school since in a public school there's a set curriculum that teachers have to follow where in private schools teachers have to make every lesson and tests. This juxtaposition between the painting and the scene of the man and woman shows the video's intent to rewrite societal norms that have been embedded for hundreds of years. While the painting shows a helpless, desperate woman who is physically below the man, the modern scene in the video demonstrates the new, equal standard for hetero relationships.

This manuscript "Computational Studies of Substrate Transport and Specificity in a Phospholipid Flippase" presents multiscale simulations to understand the details of a yeast flippase in lipid binding, membrane deformation, and protein hydration. Overall, an examination of the Drs2p-Cdc50p complex was carried out with 500-ns-long all-atom and 100-us-long coarse-grained simulations in different membrane models (pure PS, PE, PC and mixtures). Free-energy simulations were also employed to compare lipid binding free energies. A major finding is the identification of the anionic PS lipid binding to a water-filled substrate binding groove. However, I find the work lacks clarity, novelty, and biological insight.

1) My primary concern is that three different phospholipids were selected in this work: PS, PE, and PC, but only the PS lipid is anionic. First of all, it is quite obvious that the PS lipid is preferred in this limited set, due to the formal charge difference. The higher affinity of anionic lipids to transmembrane proteins has been extensively studied (too many to list, but here are a few recent examples PNAS 2020 117, 7803-7813; Structure, 2019, 27, 392-403.e3; Sci Rep. 2018, 8, 4456; Sci Rep. 2016, 6, 29502)

Second, according to prior experiments (Appl Environ Microbiol. 2014, 80, 2966-2972), the major phospholipids in yeast are phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylinositol (PI), phosphatidylserine (PS), and phosphatidic acid (PA), with minor amounts of cytidinediphosphate-diacylglycerol (CDP-DAG). There are also glycosphingolipids, ergosterol, and proteins. None of the membrane models simulated in this work is an approximate to the realistic yeast cellular membrane. Because the lipid composition has important physiological impacts, I found a lack of justification of why key anionic lipids (like PI and PA) and ergosterol were not included.

2) In addition, it was claimed "As our atomistic simulations were limited to 0.5-1.0 𝜇𝑠 due to their high computational cost". I cannot agree with the authors, given the system size of ~340,000 atoms. It is not rare to see microsecond or multiple-microsecond all-atom simulations (of this size or larger) in current studies of membrane proteins. Further, longer simulations might be more likely to sample lipid exchange and competition within the groove, as well as relevant protein conformational changes (which cannot be captured in CG simulations).

3) Moreover, while I found the results presented in Fig. 5 quite interesting, the related paragraphs seem to lack the in-depth analysis and clarity to support "a 'credit-card'-like model" First, it is not clear to me how this lipid in Fig. 5 was selected. How did this lipid look in the outer leaflet vs. in the deep state of the groove? Second, there is no analysis of the event at ~21-23 us when the lipid starts to transition. What was the trigger of the event? Were there any specific interactions? Last but not the least, as the authors said "X-ray diffraction and Cryo-EM experiments on ATP8A1 and ATP11C show density for PL head groups", it is possible to compare the simulation results (lipid density) to the experimental density. It would greatly strengthen this paper if such analysis is included.

4) The "water-filled cavities" results overall may need more clarification and probably even experimental support. First of all, how were the AA simulations compared with CG simulations, in terms of the cavities? Given the ENM constraints, there were little conformational changes of the cavities (of the protein) in response to PS moving the groove. There might be some induced fit effect and the cavities may adopt different shapes when such effect is fully considered in the AA modeling. Second, is there any experimental evidence to support this observation from MD simulations? For example, mutation of the key residue Ile508, suggested by the authors to separate the two cavities.

Thanks for the comments. We were also confused about the conflicting results which may be caused by the different tagged constructs or different concentration of plasmids used for the transfection. However, we cannot repeat the observation that Smurf1 has obvious effects on Talin-head degradation even though we performed the experiments at many different conditions and using several cell lines. Although Dr. Huang et al. found that Smurf1 induced slightly reduced levels of Talin-H as shown in Nat Cell Biol. 2009 May;11(5):624-30, Fig. S6a, we tend to think that Talin-head is not a direct target of Smurf1. If Talin-head is a genuine target of Smurf1, it should be degraded to the same extent as Kindlin-2 is degraded by Smurf1. Thus, it needs further investigation to elucidate the mechanism of Talin-head degradation.
