    1. Limestone & Karst Terrain The Valley’s abundant limestone is evidence that the area was once under ocean water, long before the tectonic plates collided. Limestone forms in warm, shallow seas when generations of tiny sea creatures die and accumulate on the ocean floor, later to compress into solid rock. Karst Because of its limestone base, the Shenandoah Valley is karst, a terrain type with distinctive landforms and water resources. Aquifers closer to the surface, used for irrigation and drinking water, are re-charged by rainwater. They are very useful but also extremely vulnerable to contamination. Caves & Caverns The water that dissolves limestone and other soluble rocks in the Shenandoah Valley leaves behind caves and caverns. In some caves and caverns, the deposits of dissolved minerals left by dripping water make the features of stalactites, stalagmites, and columns. Luray Caverns, a National Natural Landmark in the Valley, is ornately decorated with cascades, columns, stalactites, stalagmites and pools. Other Rocks & Minerals Many rocks and minerals occupy the Shenandoah Valley besides limestone. Other sedimentary rocks in the region are sandstone, shale, and coal. There are igneous rocks like granite, gabbro, and basalt, and metamorphic rocks like slate, gneiss, and quartzite. Manganese, iron ores, zinc, lead, sulfur, gypsum, and pyrite are relatively common Shenandoah Valley minerals. The Valley yields small amounts of oil and natural gas. Also present is red clay, an oceanic sediment with iron oxide, along with fossils of sea creatures like brachiopods and trilobites.

      There's a difference in productivity for it's own sake, but being both productive in the send of time spent efficiently and productive in the sense of producing something of greater value with your time than you might have spent doing something else which was less valuable, but which might still have been time well spent.

    1. The main thing I learned while reading through Phyllis Diller's jokes is that comedy has changed a lot since she started her career in the mid-1950s. Her comedy is focused on short one-liners that get laughs in quick succession, while today's comedy is more story-driven. Although a lot of her jokes are very time-bound due to their content, it was interesting to get a glimpse of what was happening at the time a joke was written. Each joke card has a date on it, and the cards span the 1960s to the 1990s. The topic of the jokes told a lot about what people were worried about or focused on at the time the joke was written, whether it was the inflation or student protests of the 1970s, a celebrity's many marriages, or gossip about the president at the time. While, like any comedian, some of her jokes fall flat, I appreciated Diller's hard work in meticulously recording, testing, and filing each joke in the gag file, along with her ability to make a joke about almost any topic.

      I love the concept of "slender ideas" as small, fleeting notes which might accumulate into something if written down. In saying "Close the notebook and think about it for a few days" Patricia Highsmith seems to be suggesting that one engage in diffuse thinking, passive digesting, or mulling rather than active or proactive thinking.

      She also invokes the magic word "presto!" (which she exclaims) as if to indicate that magically the difficult work of writing is somehow no longer difficult. Many writers seem to indicate that this is a phenomenon, but never seem to put their finger on the mechanism of why it happens. Some seems to stem from the passive digestion over days with diffuse thinking, with portions may also stem from not starting from a blank page and having some material to work against instead of a vacuum.

      This is a book about a box that contained the world. The box was the Picture Academy for the Young, a popular encyclopedia in pictures invented by preacher-turned-publisher Johann Siegmund Stoy in eighteenth-century Germany. Children were expected to cut out the pictures from the Academy, glue them onto cards, and arrange those cards in ordered compartments—the whole world filed in a box of images.

      As Anke te Heesen demonstrates, Stoy and his world in a box epitomized the Enlightenment concern with the creation and maintenance of an appropriate moral, intellectual, and social order. The box, and its images from nature, myth, and biblical history, were intended to teach children how to collect, store, and order knowledge. te Heesen compares the Academy with other aspects of Enlightenment material culture, such as commercial warehouses and natural history cabinets, to show how the kinds of collecting and ordering practices taught by the Academy shaped both the developing middle class in Germany and Enlightenment thought. The World in a Box, illustrated with a multitude of images of and from Stoy's Academy, offers a glimpse into a time when it was believed that knowledge could be contained and controlled.

    1. The box makes me feel connected to a project. It is my soil. I feel this evenwhen I’ve back-burnered a project: I may have put the box away on a shelf, but Iknow it’s there. The project name on the box in bold black lettering is a constantreminder that I had an idea once and may come back to it very soon.

      Having a physical note taking system also stands as a physical reminder and representation of one's work and focus. It may be somewhat out of the way on a shelf, but it takes up space in a way that digital files and notes do not. This invites one into using and maintaining it.

    1. The bookitself participates in the history it recounts: it has a title page, table of contents,footnotes, a bibliography and an index to assist the reader, while the digitalcopy enables the reader to search for individual words and phrases as well asto copy-and-paste without disfiguring a material object.

      Some scholars study annotations as part of material culture. Are they leaving out too much by solely studying those physically left in the books about which they were made, or should we instead also be looking at other sources like commonplace books, notebooks, note cards, digital spaces like e-readers that allow annotation, social media where texts are discussed, or even digital marginalia in services like Hypothes.is or Perusall?

      Some of these forms of annotation allow a digital version of cut and paste which doesn't cause damage to the original text, which should be thought of as a good thing though it may separate the annotations from the original physical object.

      is a good solution since it defeats the "h1 cannot be a child of h1" DOM error + let us use the sx prop.

      We would not be able to use sx prop to style the inline part if we would use the native <span> component ie. <Typography > yo mama <span> so fat </span> </Typography>

      You would have to use inline style styling instead.

      This <Box/> method is the preffered way of doing it. The <Paper/> method too is a good idea. The basic reasoning is that only MUI components support the sx prop ie. only they can fully use the MUI systems features. Thus, it doesnt make sense to use the native <img/> component in the markup, as it does not support sx

    1. The SX prop does not work with non MUI components eg. native <img/> component. To style this, you will have to use the native inline styling ie. <img style=" ............". /> If you want to style the img and use some MUI system specific features, ie. you need to use sx ; do the following. Use <Box component="img" sx={{ }} /> instead. This supports the sx prop, thus your image will be customizable with sx prop.

    1. Color

      The color prop does not support all MUI theme colors / their shades DIRECTLY through its color prop. Eg. The color prop supports primary but not primary.main/dark. It does not support the palette warning color at all.

      To remedy this you have 2 options 1 BETTER -- give the icon the desired color through the sx prop OR 2. wrap the icon in a box and give the box to desired color

    1. Honestly, I don't know. Strictly speaking it's different to a window being opened at a specific size (I can see the difference). I don't know whether that difference is material. Seeing as these are tests, I'd like to remove as many unknowns as possible. If someone can tell me that it makes no difference then I'd be happy.

This article isn’t to argue whether it should be this way or not, though; it’s just to change yours such that your MenuItem(s) show below the menu selection, like so:

    1. Customization of the TextField can be cumbersome with the classes API, where you have to define the the classes prop. It's easier to use the default values, as described above. For example:

      It's surprising that they show an example using styled-components, which is a competing style library to their own @material-ui/styles, and even admit that this might be preferred over using the classes API, which they admit may be cumbersome.

      I like that they are open-minded enough to acknowledge that there are cases where built-in API may be too cumbersome, and even show examples of using styled-components.

    1. Demonstrates how label text will wrap at a point that appears to narrow when shrunk (the label can't even be as wide as the input it is labeling!), and how to work around this problem by adding styles:

      Of course, you would only want to do this if you are going to only be showing the label in shrunk state (which I think is safe to say is the case for date picker inputs), since it would look bad to actually have text overflowing outside of the input box.       Reference

      The input label "shrink" state isn't always correct. The input label is supposed to shrink as soon as the input is displaying something. In some circumstances, we can't determine the "shrink" state (number input, datetime input, Stripe input). You might notice an overlap.

      To workaround the issue, you can force the "shrink" state of the label.

      You need to make sure that the input is larger than the label to display correctly.

Transformation of calcium-competent cells was carried out by the procedure detailed below: •The competent bacterial cells were thawed briefly and 200 μL of cells was mixed rapidly with plasmid DNA (10-50 ng) in fresh, sterile microcentrifuge tubes and maintained on ice for 30 min. A negative control with competent cells only (no added DNA) was also included. •Cell membranes were disrupted by subjecting cells to heat-pulse (42 °C) for 90 sec. •After heat shock, cells were incubated on ice for 5 min. •Cells were then mixed with 1 mL LB medium and incubated with shaking at 37 °C for 1 h. •For blue/white screening 40 μL of X-gal solution (20 mg mL-1 in dimethylformamide) and 4 μL of the IPTG (200 mg mL-1) was spread on LB-ampicillin (LB-amp) plates with a sterile glass rod. The plate was allowed to dry for 1h at 37 °C prior to spreading of bacterial cells. •Bacterial cells (100-200 μL) were spread and the plate was incubated at 37 °C for overnight. •White colonies were picked from the plates and suspended into LB-amp broth and cultivated to OD600=0.5
    2. 2 mL of an overnight culture of E. coli cells was inoculated into 100 mL LB medium and incubated with vigorous shaking at 30 °C until A600 of 0.8 was reached. •Cells were collected in 50 mL plastic (Falcon) tubes, cooled for 15 min on ice and centrifuged in a pre-cooled centrifuge (4,000 rpm for 10 min at 4 °C). •The pellet was suspended in 20 mL of ice-cold 50 mM CaCl2-15% glycerol solution, maintained on ice for 15 min and centrifuged again at 4,000 rpm for 10 min at 4 °C. •Pellet was resuspended in 2 mL of ice-cold 50 mM CaCl2-15 % glycerol solution, kept on ice for 30 min and aliquoted in 400 μL in microcentrifuge tubes. These were stored at -80 °C until required.
    3. Preparation of electrocompetent cells (E. coli cells) A protocol was employed. The procedure was carried out in cold under sterile conditions as follows: •A single colony of E. coli DH10B/ DH5α/XL1blue was inoculated in 20 mL of LB medium and grown overnight at 30 °C. •500 mL LB medium was inoculated with 5mL of this overnight grown culture of the E. coli and incubated with vigorous shaking (250 rpm) at 30 °C until an A600of 0.5 - 0.8 was achieved. •The cells were chilled in ice for 10-15 min and transferred to prechilled Sorvall® centrifuge tubes and sedimented at 4,000 rpm for 20 min at 4 °C. •The supernatant was decanted and cells were resuspended in 500 mL of sterile ice-cold water, mixed well and centrifuged as described above. •The washing of the cells described above was repeated with 250 mL of sterile ice-cold water, following which cells were washed with 40 mL of ice-cold 10 % (v/v) glycerol and centrifuged at 4,000 rpm for 10 min. •The glycerol solution was decanted and the cell volume was recorded. The cells were resuspended in an equal volume of ice-cold 10 % glycerol. •Cells were then dispensed in 40 μL volumes and stored at -80 °C until required.
    4. In order to minimize self ligation of vector during cloning experiments, the digested DNA was subsequently treated with calf intestinal phosphatase (CIP) [NEB, UK]. The reaction conditions and amount of CIP were optimized and varied from (0.06-1) unit/picomole DNA termini. The dephosphorylation reaction was carried out in 50 μL reaction as follows. Reaction mixture containing no restriction enzyme was treated as control. Reaction was incubated for 1 h at 37 °C and stopped by heat inactivation at 65 °C for 20 min. 2.5.5. Composition of restriction mixture (50 μL) Linearized Plasmid DNA X μL (1 μg) CIP 1 μL (0.06-1 U μL-1) Reaction buffer (10X) 5.0 μL Distilled water Y μL Total volume 50 μL Linearized and dephosphorylated plasmids from each reaction were purified from low melting agarose gel using gel extraction method according to the manufacturer’s protocol (Qiagen gel extraction kit, Germany). 100 ng DNA from each reaction was then ligated in15 μL reaction volume containing 1.5 μL of 10X ligation buffer (NEB, England) and 0.2 μL of T4 DNA ligase to check the efficiency of self ligation after dephosphoryaltion. The ligation mixture was incubated at 16 °C for overnight and transformed into E. coli DH5αcompetent cells.
    5. The isolated DNA was diluted (1:100) with MQ. The concentration (mg mL-1) of the DNA [N] was determined spectrophotometrically by recording absorbance at 260 nm (A260) as: A260 = ε 260[N]where ε 260 is the extinction coefficient of DNA (50 for ds DNA) [N] = concentration (mg mL-1) of DNA The concentration of ds DNA [N] was calculated as [DNA] (mg mL-1) = A260/ε 260 [DNA] (μg mL-1) = A260 × 50 × dilution factor Purity of DNA was checked by measuring absorbance at 260 and 280 nm and calculating the A260/A280 ratio (Sambrook et al., 1989). A DNA sample was considered pure when A260/A280 ranged between 1.8-1.9. An A260/A280 < 1.7 indicated contamination of the DNA preparation with protein or aromatic substances such as phenol, while an A260/A230 < 2.0 indicated possible contamination of high molecular weight polyphenolic compounds like humic substances.
    6. Determination of DNA quantity and purity
    7. as well as commercial methods (MN kit, Germany; Mo-Bio kit, CA, USA; Zymo soil DNA kit, CA, USA) according to the manufacturer’s protocols and compared in terms of DNA yield and purity.

Soil (1 gm) was suspended with 0.4 gm (w/w) polyactivated charcoal (Datta and Madamwar, 2006) and 20 μL proteinase K (10 mg mL-1) in 2 mL of modified extraction buffer [N,N,N,N cetyltrimethylammonium bromide (CTAB) 1% w/v, polyvinylpolypyrrolidone (PVPP) 2% w/v, 1.5 M NaCl, 100mM EDTA, 0.1 M TE buffer (pH 8.0), 0.1M sodium phosphate buffer (pH 8.0) and 100 μL RNaseA] [Zhou et al., 1996] in 20 mL centrifuge tubes to homogenize the sample and incubated at 37 °C for 15 min in an incubator shaker at 200 rpm. Subsequently, 200 μL of 10% SDS was added to the homogenate and kept at 60 °C for 2 h with intermittent shaking. DNA was precipitated by adding 0.5 V PEG 8000 (30 % in 1.6 M NaCl) and left at room temperature for an hour (Yeates et al., 1998). The precipitated DNA was collected by centrifugation at 8000 x g at 4 °C. The supernatant was discarded and pellet was dissolved in 1 mL of TE buffer (pH 8.0) and then100 μL of 5 M potassium acetate (pH 4.5) was added and incubated at 4 °C for 15 min. The supernatant was collected after centrifugation at 8000 x g and treated with equal volumes of phenol: chloroform (1:1) followed by chloroform: isoamylalcohol (24:1) at 8000 x g for 15 min
